Calsequestrin-1 (CASQ1) is the main calcium-buffering protein in the sarcoplasmic reticulum of skeletal muscle. It is required for excitation-contraction coupling. Our antibodies are the ideal research tool for the study of muscle diseases such as malignant hyperthermia and myopathies, as well as fundamental aspects of muscle function.

CASQ1: The Calcium Reservoir of Skeletal Muscle

Calsequestrin-1 (CASQ1) is a high-capacity, moderate-affinity calcium-binding protein that resides in the terminal cisternae of the sarcoplasmic reticulum (SR) of skeletal muscle. The protein serves as a calcium sink to facilitate storage of the massive amounts of calcium needed for muscle contraction at very low free calcium concentrations. The protein also likely modulates activity of the ryanodine receptor calcium release channel through polymerization that is dependent on calcium. Mutations in CASQ1 cause vacuolar aggregate myopathy and predispose patients to malignant hyperthermia, a life-threatening reaction to certain anesthetics.

Background

The protein encoded by this gene is the skeletal muscle specific member of the calsequestrin protein family. Calsequestrin is a luminal sarcoplasmic reticulum calcium sensor in cardiac and skeletal muscle cells. The protein encoded by this gene, also known as calmitine, is a calcium regulator of mitochondria in skeletal muscle. This protein is absent in the Duchenne and Becker type of muscular dystrophy.
