CBS: A Master Regulator of Sulfur Metabolism

Cystathionine Beta-Synthase (CBS) is a pyridoxal phosphate (vitamin B6)-dependent enzyme that catalyzes the first step in the transsulfuration pathway, the condensation of homocysteine and serine to form cystathionine. This pathway accounts for the majority of homocysteine clearance, a toxic metabolite, and is the predominant source of the gaseous signaling molecule hydrogen sulfide (H2S) in the liver and brain. Homocystinuria, a severe inherited metabolic disease caused by loss-of-function mutations in the CBS gene is defined by an increase in plasma homocysteine leading to intellectual disability, skeletal deformities, and vascular disease.

Background

The protein encoded by this gene acts as a homotetramer to catalyze the conversion of homocysteine to cystathionine, the first step in the transsulfuration pathway. The encoded protein is allosterically activated by adenosyl-methionine and uses pyridoxal phosphate as a cofactor. Defects in this gene can cause cystathionine beta-synthase deficiency (CBSD), which can lead to homocystinuria. This gene is a major contributor to cellular hydrogen sulfide production. Multiple alternatively spliced transcript variants have been found for this gene.
