CDC7: The Initiator of DNA Replication

Cell Division Cycle 7-Related Protein Kinase (CDC7), commonly referred to as CDC7, is a serine/threonine kinase that is critical for the initiation of DNA replication. It is tightly regulated during the cell cycle, with its activity peaking at the G1/S transition. Its primary function is to phosphorylate a number of the subunits of the MCM (minichromosome maintenance) complex, which is the core helicase at replication origins. This is the “firing” signal that activates the MCM helicase, which loads the DNA replication machinery and initiates synthesis. Many cancer cells are under replicative stress, and thus are particularly sensitive to the inhibition of CDC7, making it an attractive target.
