CDK7: The Master Kinase of Transcription and Cell Cycle
Cyclin-Dependent Kinase 7 (CDK7) is a serine/threonine kinase with two related but distinct functions in the cell. First, as the catalytic subunit of the CDK-Activating Kinase (CAK) complex, CDK7 serves as the global on/off switch for cell cycle progression. Second, and most importantly, CDK7 is also a subunit of the Transcription Factor II H (TFIIH) complex, where it phosphorylates the C-Terminal Domain (CTD) of RNA Polymerase II. This is the essential step that releases Pol II from the promoter for productive elongation. As CDK7 acts as the global gatekeeper for transcription across the entire genome, it is an effective way to take down the entire oncogenic network.

Background
The protein encoded by this gene is a member of the cyclin dependent protein kinase (CDK) family. Members of the CDK family are closely related to the gene products of the Saccharomyces cerevisiae cdc28 and Schizosaccharomyces pombe cdc2 and have been shown to be important regulators of cell cycle progression. This protein is part of a trimeric complex with cyclin H and MAT1 that functions as a Cdk-activating kinase (CAK). It is an essential component of the transcription factor TFIIH, which participates in transcription initiation and DNA repair. This protein may provide a direct link between transcription regulation and the cell cycle.
