CIB2: An Essential Calcium-Binding Protein for Hearing
Calcium and integrin binding family member 2 (CIB2) belongs to a family of EF-hand calcium-binding proteins. It is abundantly expressed in the stereocilia of sensory hair cells of the inner ear and in retinal photoreceptors. CIB2 is a constituent of the mechano-electrical transduction (MET) complex in the hair cells of the inner ear. The MET complex is the protein machinery that translates sound vibrations into a neural signal. CIB2 likely acts as a calcium sensor or buffer in the MET complex, playing a role in adaptation and sensitivity regulation. Mutations in this gene are known to cause non-syndromic deafness (DFNB48) and a type of Usher syndrome (USH1J). Usher syndrome is a tragic disorder leading to deafness and progressive blindness.

Background
The protein encoded by this gene is homologous to KIP/CIB, calcineurin B, and calmodulin. The gene product is a calcium-binding regulatory protein, which binds to DNA-dependent protein kinase catalytic subunits (DNA-PKcs), with a role in photoreceptor cell maintenance. Mutations in this gene cause deafness, autosomal recessive, 48 (DFNB48), and also Usher syndrome 1J (USH1J). Alternative splicing results in multiple transcript variants.
