CLASP1: A Key Regulator of Microtubule Dynamics

Cytoplasmic Linker Associated Protein 1 (CLASP1) is a “plus-end tracking protein” (+TIP) that associates with the growing end of microtubules. It plays a vital role in stabilizing dynamic microtubules by promoting microtubule rescue (switching from shrinkage to growth) and anchoring the microtubules to specific sites, such as the cell cortex and chromosome kinetochores during mitosis. This activity is crucial for the formation of a functional mitotic spindle, chromosome segregation, and the establishment of cell polarity during cell migration. The protein is frequently dysregulated in cancers and is linked to genomic instability and increased metastatic potential.

Background

CLASPs, such as CLASP1, are nonmotor microtubule-associated proteins that interact with CLIPs (e.g., CLIP170; MIM 179838). CLASP1 is involved in the regulation of microtubule dynamics at the kinetochore and throughout the spindle.
