CLPS: The Essential Cofactor for Fat Digestion

CLPS is a small protein that is produced in the pancreas, and secreted into the small intestine in an inactive pro-form. In the intestine, CLPS is activated by proteolytic cleavage of proCLPS by trypsin. Colipase has a single function, to serve as a non-enzymatic cofactor for pancreatic lipase. Bile salts present in the gut coat fat droplets, and prevent the lipase enzyme from binding to the triglycerides it is meant to digest. Colipase binds both to the fat droplet and to the lipase, thus tethering the lipase enzyme to its substrate, where it can act to efficiently digest triglycerides into absorbable fatty acids. Colipase is therefore essential for the digestion of dietary fats.

Background

Colipase is a cofactor of pancreatic lipase. It allows the lipase to anchor itself to the lipid-water interface. Without colipase the enzyme is washed off by bile salts, which have an inhibitory effect on the lipase.
