CPA6: An Extracellular Matrix-Bound Peptidase in the Brain

Carboxypeptidase A6 (CPA6) is part of the A/B subfamily of M14 metallocarboxypeptidases. It is almost exclusively expressed in the brain, where it is localized to the ECM. It is proposed to cleave off C-terminal residues of neuropeptides and other signaling ligands in the ECM to tune their activity. The modulation of peptide activity by CPA6 in this way allows for localized control of peptide signaling, which is important for neuron migration and development. Mutations in the CPA6 gene have been associated with genetic epilepsy and intellectual disability.

May be involved in the proteolytic inactivation of enkephalins and neurotensin in some brain areas. May convert inactive angiotensin I into the biologically active angiotensin II.
