CPB2: The Thrombin-Activatable Fibrinolysis Inhibitor
CPB2 is a plasma zymogen and more commonly known as the Thrombin-Activatable Fibrinolysis Inhibitor (TAFI). It is produced in the liver and secreted in an inactive form in the plasma. In blood coagulation, CPB2 is activated by the thrombin-thrombomodulin complex. The active enzyme (TAFIa) downregulates fibrinolysis (breakdown of blood clot) by removing the C-terminal lysine and arginine residues from the partially degraded fibrin, eliminating the binding sites for plasminogen, thereby preventing the clot from getting lysed. The increased level or activity of TAFI is a risk factor for thrombotic diseases such as deep vein thrombosis and myocardial infarction.

Background
Carboxypeptidases hydrolyze C-terminal peptide bonds. They are subclassified into metallo-, serine, and cysteine carboxypeptidases based on their catalytic mechanisms. They are further named based on their substrate specificity as carboxypeptidase A (aliphatic residues) or carboxypeptidase B (basic amino residues). The protein encoded by this gene is activated by trypsin and is specific for carboxypeptidase B substrates. The mature protein resulting from thrombin activation downregulates fibrinolysis. Polymorphisms have been described for this gene and for the promoter region. Alternate splicing generates multiple transcript variants.
