CPD: A Key Peptide Processing Enzyme

CPD is a metallocarboxypeptidase (enzymes that use a divalent metal cation as a cofactor for catalysis). CPD is predominantly localized to the trans-Golgi network (TGN) and secretory vesicles, and shuttles between these compartments and the plasma membrane. CPD cleaves C-terminal arginine and lysine residues from peptide and protein precursors. This enzyme is important for the post-translational processing and maturation of a wide variety of bioactive molecules, including peptide hormones, neuropeptides, and growth factors. As CPD controls the final step in their synthesis, these diverse molecules and the many physiological processes they regulate are tightly dependent on this enzyme.

Background

The metallocarboxypeptidase family of enzymes is divided into 2 subfamilies based on sequence similarities. The pancreatic carboxypeptidase-like and the regulatory B-type carboxypeptidase subfamilies. Carboxypeptidase D has been identified as a regulatory B-type carboxypeptidase. CPD is a homolog of duck gp180, a hepatitis B virus-binding protein. Transcript variants utilizing alternative polyadenylation signals exist for this gene.
