CRABP2: A Nuclear Delivery Vehicle for Retinoic Acid
Cellular Retinoic Acid-Binding Protein 2 (CRABP2) is a small cytosolic protein that, similar to its CRABP1 homolog, binds all-trans-retinoic acid (atRA) with high affinity. However, CRABP2 has a unique and opposing function. While CRABP1 facilitates atRA catabolism, CRABP2 is a shuttle. Binding atRA induces a conformational shift in CRABP2, which allows it to relocate to the nucleus. Once in the nucleus, CRABP2 directly binds the retinoic acid receptors (RARs), and then “channels” its atRA ligand into the receptor’s ligand-binding pocket. This greatly accelerates the efficiency of RAR-dependent gene transcription, establishing CRABP2 as a major positive regulator of the retinoid signaling pathway.
