CTDSP1: A Key Regulator of RNA Polymerase II
C-Terminal Domain Small Phosphatase 1 (CTDSP1), also known as CSRP1 and RPP30, is a nuclear phosphatase from the FCP1 family. Its main substrate is the C-terminal domain (CTD) of the largest subunit of RNA Polymerase II. The CTD of RNA Polymerase II consists of 52 repeats of a heptapeptide sequence in mammals. The phosphorylation state of the CTD acts as a dynamic code that regulates the entire transcription cycle. CTDSP1 specifically dephosphorylates serine-5 of these repeats. CTDSP1 has been suggested to be important for promoting transcription termination and recycling the polymerase for a new round of transcription, however its exact function is still not well understood.
