CTSG: A Potent Neutrophil Serine Protease

Cathepsin G (CTSG) is a chymotrypsin-like serine protease that is secreted by neutrophils into their azurophilic granules. It is released into the extracellular space upon activation of neutrophils during an inflammatory response. Cathepsin G is a proteolytic enzyme that has robust activity against various extracellular matrix components, including elastin and fibronectin. Cathepsin G can be protective by killing invading pathogens during an immune response. However, unregulated or excessive Cathepsin G activity can be detrimental, as it is a major cause of the tissue damage that is a hallmark of many chronic inflammatory diseases, such as chronic obstructive pulmonary disease (COPD), rheumatoid arthritis, and cystic fibrosis.

Background

This gene encodes a member of the peptidase S1 (chymotrypsin) family of serine endopeptidases. The encoded preproprotein is proteolytically processed to generate a mature protein product. This product appears to play a role in host defense, including inflammation and antigen processing. Homozygous knockout mice for this gene exhibit enhanced susceptibility to bacterial and fungal infection.
