CYP2C8: A Major Metabolizer of Lipids and Taxanes
Cytochrome P450 2C8 (CYP2C8) is a monooxygenase enzyme expressed in liver and small intestine. As one of the principal CYP450 enzymes, CYP2C8 is a major catalyst for the phase I metabolism of a number of clinically used drugs, particularly the anti-cancer taxane drugs (paclitaxel, docetaxel) and lipid lowering statin drugs (cerivastatin). In endogenous lipid metabolism, CYP2C8 generates epoxyeicosatrienoic acids (EETs) from arachidonic acid, which have potent vasodilator and anti-inflammatory activity. CYP2C8 is widely distributed in body tissues, and due to its extensive substrate range, plays a key role in determining both drug toxicity and drug efficacy.

Background
Cytochromes P450 are a group of heme-thiolate monooxygenases. In liver microsomes, this enzyme is involved in an NADPH-dependent electron transport pathway. It oxidizes a variety of structurally unrelated compounds, including steroids, fatty acids, and xenobiotics. In the epoxidation of arachidonic acid it generates only 14,15- and 11,12-cis-epoxyeicosatrienoic acids. It is the principal enzyme responsible for the metabolism the anti-cancer drug paclitaxel (taxol).
