CYP2C9: The Major Metabolizer of Warfarin
Cytochrome P450 2C9 (CYP2C9) is the most abundant monooxygenase enzyme in the CYP2C subfamily, and it is the single most important enzyme responsible for metabolizing a wide range of clinically used drugs. CYP2C9 metabolizes around 16% of all drugs, including the anticoagulant warfarin, numerous NSAIDs (such as ibuprofen), and oral hypoglycemics. This enzyme has high genetic polymorphism, with patients having low-activity variants requiring much lower dosages of drugs like warfarin in order to avoid life-threatening bleeding. CYP2C9 pharmacogenomics is, therefore, a critical component of personalized medicine.
