CYP3A7: The Fetal Drug Metabolizer

Cytochrome P450 3A7 (CYP3A7) is an endoplasmic reticulum-based monooxygenase enzyme, and is the main isoform of the CYP3A family found in human fetal liver. Expression of this gene is rapidly downregulated at birth, and is replaced by expression of the adult isoform, CYP3A4. CYP3A7 is responsible for the metabolism of many fetal steroids as well as maternal drugs that can pass through the placental barrier. The gene is silenced in the adult liver, but can be aberrantly re-expressed in certain malignancies (such as hepatocellular carcinoma) where it is associated with a fetal metabolic phenotype.
