DNPEP: A Key Regulator of Angiotensin Peptides in the Brain

DNPEP, or Aspartyl Aminopeptidase, is a metallopeptidase enzyme that cleaves N-terminal aspartate and glutamate residues from peptides and proteins. DNPEP's best-known substrate is angiotensin II, as it acts as the key regulator of angiotensin peptides in the brain. It metabolizes angiotensin II to angiotensin III. This conversion is an important regulator of the brain renin-angiotensin system (RAS) and can have profound effects on the cardiovascular system. Angiotensin III is a major effector of the central actions of RAS, including cardiovascular and water homeostasis, and DNPEP activity can impact this signaling. Thus, this enzyme is an attractive, novel target for hypertension and related disorders.

Background

The protein encoded by this gene is an aminopeptidase which prefers acidic amino acids, and specifically favors aspartic acid over glutamic acid. It is thought to be a cytosolic protein involved in general metabolism of intracellular proteins.
