Background
Fas/FasL-mediated apoptosis is one of the important mechanisms of gastric cancer development. Fas-associated factor 1 (FAF1) and Fas-inhibitory protein (FLIP) are in Fas/ The FasL-mediated apoptotic pathway acts on the same site to promote and inhibit apoptosis, respectively. Fas1 is a protein discovered in 1995 that enhances the induction of apoptosis. It is suggested that the amino acid sequence of FAF1 at position 181-381 can interact with the DED of FADD and caspase-8, and also constitute the composition of Fas death-inducing signaling complex. Partially binds to FADD and caspase-8, enhances mediated apoptosis, and has the specific low expression in gastric cancer tissues. Fas-associated factor 1 (FAF1) is a pro-apoptotic protein that induces Fas-mediated apoptosis. FAF1 is a member of the newly discovered Fas death signal conjugate, and its N-terminal region can specifically bind to Fas, which is a protein molecule capable of enhancing Fas-mediated apoptosis. It encodes a protein involved in the survival of tumor cells and the onset of some tumors in humans. Even in the absence of any exogenous death signals, FAF1 overexpression initiates apoptosis. In addition to interacting with Fas, FAFI1 is also a member of the regulatory subunit of protein kinase CK2, which binds to each other as a complex, and its induction of apoptosis is significantly increased when stimulated by DNA damage.
