HTR2C: A Key Therapeutic Target in Metabolic and Neuropsychiatric Disorders
HTR2C, a serotonin receptor subtype, plays a crucial role in regulating appetite, mood, and metabolic processes. Dysfunction contributes to obesity and type 2 diabetes through impaired satiety signaling and glucose homeostasis, with selective agonists demonstrating appetite-suppressing effects in clinical trials. In psychiatric disorders, genetic variants associate with suicide risk and treatment-resistant depression, potentially influencing SSRI efficacy through downstream signaling pathways. The receptor's involvement in dopamine modulation links it to antipsychotic drug actions, though its activation can paradoxically exacerbate metabolic side effects. Premenstrual dysphoric disorder symptoms correlate with hormonal regulation of receptor expression patterns. Recent evidence suggests roles in addiction through reward pathway modulation, particularly in nicotine and cocaine dependence. Autoimmune conditions like Sjögren's syndrome show autoantibody interactions that may affect secretory functions. Therapeutic targeting remains complex due to opposing effects in different tissues, with ongoing development of biased ligands to separate metabolic benefits from psychiatric side effects. Polymorphisms influence individual drug responses, highlighting its pharmacogenomic importance in personalized medicine approaches for both neurological and metabolic conditions.

Background
This gene encodes a seven-transmembrane G-protein-coupled receptor. The encoded protein responds to signaling through the neurotransmitter serotonin. The mRNA of this gene is subject to multiple RNA editing events, where adenosine residues encoded by the genome are converted to inosines. RNA editing is predicted to alter the structure of the second intracellular loop, thereby generating alternate protein forms with decreased ability to interact with G proteins. Abnormalities in RNA editing of this gene have been detected in victims of suicide that suffer from depression. In addition, naturally-occuring variation in the promoter and 5' non-coding and coding regions of this gene may show statistically-significant association with mental illness and behavioral disorders.
