IGF2R: The Dual-Function IGF2 and M6P Receptor
The IGF2R is a large, single-pass transmembrane protein that has two distinct and critical functions. First, it is a clearance receptor for the potent mitogen, insulin-like growth factor 2 (IGF2). Binding extracellular IGF2 and internalizing it for lysosomal degradation, IGF2R acts as a tumor suppressor by sequestering mitogenic IGF2 and preventing its signaling through IGF1R. Second, IGF2R is also the Cation-Independent Mannose-6-Phosphate Receptor (CI-M6PR) and is the major lysosomal enzyme trafficking receptor that shuttles newly synthesized mannose-6-phosphate-tagged enzymes from the Golgi to the lysosome.

Background
IGF2R is a gene that encodes a receptor for the insulin-like growth factor 2 (IGF2), a protein involved in growth and development. The IGF2R receptor primarily functions by clearing IGF2 from the bloodstream, regulating its levels, and preventing overactivation of IGF2-mediated signaling. This is important for controlling cellular growth and differentiation, as dysregulation can lead to abnormal growth or contribute to cancer development. IGF2R is also involved in cellular processes such as endocytosis and is implicated in diseases like cancer, particularly in the regulation of tumor growth and metastasis.
