MTHFD2: A Mitochondrial Engine for Cancer Cell Proliferation

Methylenetetrahydrofolate Dehydrogenase 2 (MTHFD2) is a mitochondrial enzyme involved in one-carbon metabolism that catalyzes two consecutive reactions of the folate cycle. MTHFD2 is a housekeeping gene that is highly expressed during embryogenesis but is silenced in most adult differentiated tissues. By contrast, MTHFD2 is hyper-expressed in a broad range of cancers, where it fuels the biosynthesis of nucleotides and other macromolecules required for the proliferation of cancer cells. The highly cancer-specific expression profile of MTHFD2 makes it an ideal and highly druggable target for next-generation cancer therapeutics.

Background

The protein encoded by this gene is a nuclear-encoded mitochondrial bifunctional enzyme with methylenetetrahydrofolate dehydrogenase and methenyltetrahydrofolate cyclohydrolase activities. This enzyme is a homodimer. It is distinct in having an absolute requirement for magnesium and inorganic phosphate. Binding of magnesium is necessary for the enzyme-NAD complex to form. Two different transcripts have been described as a result of alternative splicing, one protein-coding and one not protein-coding. There is a pseudogene of this gene on chromosome 7.
