NARS: The Cytoplasmic Gatekeeper of Asparagine in Protein Synthesis
Asparaginyl-tRNA Synthetase 1 (NARS1) is a housekeeping enzyme localized to the cytoplasm. It is a member of the aminoacyl-tRNA synthetases family. Aminoacyl-tRNA synthetases are responsible for the first step of protein synthesis. NARS1 recognizes and catalyzes the ligation of asparagine and its cognate transfer RNA (tRNA-Asn) in an ATP-dependent manner. This reaction "charges" tRNA-Asn with asparagine, which is a prerequisite for the amino acid's incorporation into growing polypeptide chains in the process of translation. Bi-allelic mutations in the NARS1 gene abrogate this crucial function, causing rare neurodevelopmental diseases.
