NAT1: A Key Player in Drug Metabolism and Carcinogenesis
N-acetyltransferase 1 (NAT1) is a cytosolic enzyme that acetylates arylamine and heterocyclic amine substrates. NAT1 is involved in metabolism of many therapeutic drugs and is also responsible for detoxification of environmental carcinogens. In some cases, NAT1 bioactivates pro-carcinogens, which makes it a risk factor for carcinogenesis in various tissues such as the bladder and colon. Polymorphisms of the NAT1 gene give rise to fast, slow, and intermediate acetylator phenotypes which significantly affect therapeutic efficacy and susceptibility to toxicity in affected individuals. NAT1 is therefore an important pharmacogenomic biomarker and an active area of personalized medicine.

Background
This gene is one of two arylamine N-acetyltransferase (NAT) genes in the human genome, and is orthologous to the mouse and rat Nat2 genes. The enzyme encoded by this gene catalyzes the transfer of an acetyl group from acetyl-CoA to various arylamine and hydrazine substrates. This enzyme helps metabolize drugs and other xenobiotics, and functions in folate catabolism. Multiple transcript variants encoding different isoforms have been found for this gene.
