NDST1: A Key Modifying Enzyme in Heparan Sulfate Biosynthesis

Located in the Golgi apparatus, N-deacetylase and N-sulfotransferase 1 (NDST1) is a bifunctional enzyme responsible for an early modification step during the biosynthesis of heparan sulfate proteoglycans (HSPGs). This essential sulfotransferase first removes the acetyl group from N-acetylglucosamine residues and then transfers a sulfate group in its place. The overall structure of the heparan sulfate chain is crucial to its ability to bind and regulate the activities of many signaling molecules, including growth factors (FGF and HGF), chemokines and morphogens. Therefore, NDST1 functions as a master regulator of development, angiogenesis and inflammation.

Background

The gene encodes a member of the heparan sulfate/heparin GlcNAc N-deacetylase/ N-sulfotransferase family. The encoded enzyme is a type II transmembrane protein that is localized in the Golgi apparatus. The encoded protein transfers sulfate from 3'-phosphoadenosine 5'-phosphosulfate to the nitrogen of glucosamine in heparan sulfate. Alternative splicing results in multiple transcript variants.
