NT5M: A Pivotal Regulator in Metabolic Disorders Cancer Resistance and Neurological Diseases

NT5M, a key enzyme in nucleotide metabolism, plays a critical role in maintaining cellular homeostasis and has been implicated in various disease pathways. As a cytosolic 5'-nucleotidase, it regulates purine and pyrimidine levels by hydrolyzing nucleoside monophosphates, influencing DNA repair, replication, and energy metabolism. Dysregulation of this enzyme disrupts nucleotide balance, contributing to metabolic disorders such as mitochondrial dysfunction and impaired energy production. In cancer, aberrant activity is linked to chemoresistance, as altered nucleotide pools enable tumor cells to evade therapy-induced DNA damage. Evidence also suggests involvement in cardiovascular diseases, where disrupted purine metabolism exacerbates ischemic injury and oxidative stress. Neurological conditions, including neurodegenerative disorders, may arise from impaired nucleotide recycling, leading to synaptic dysfunction and neuronal death. Additionally, its role in immune modulation highlights potential connections to autoimmune and inflammatory diseases. Given its broad impact on cellular processes, targeting this pathway offers therapeutic potential for conditions characterized by metabolic imbalance, genomic instability, or aberrant proliferation. Research continues to explore its mechanisms in disease progression and treatment resistance.

Background

This gene encodes a 5 nucleotidase that localizes to the mitochondrial matrix. This enzyme dephosphorylates the 5- and 2(3)-phosphates of uracil and thymine deoxyribonucleotides. The gene is located within the Smith-Magenis syndrome region on chromosome 17.

Recombinant Mouse anti-Human NT5M Monoclonal antibody (EML1700) (CAT#: MOB-1470CT)
Recombinant Mouse anti-Human NT5M works well in WB for detecting endogenous protein in HEK293 and HepG2 lysates. Tested in human liver and kidney tissues, it shows clean bands with minimal background when using 3% BSA blocking. While it performs consistently in overexpression systems, detection in low-expression samples may require optimization. No cross-reactivity observed with related nucleotidases. A reliable option for basic research applications.

Recombinant Mouse anti-Human NT5M Monoclonal antibody (EML1700) (CAT#: MOB-1470CT)
The EML1700 anti-NT5M antibody has proven useful for comparative studies across different cell lines. In our hands, it consistently detected the target in both normal and cancer cell lysates with good specificity. The antibody worked particularly well for monitoring NT5M expression changes under metabolic stress conditions. While the signal intensity is moderate rather than exceptionally strong, we appreciated the clean background when using recommended protocols. Storage stability appears good after several freeze-thaw cycles.
