OGDH: A Central Metabolic Regulator in Neurodegeneration and Cancer

OGDH is a critical mitochondrial enzyme in the tricarboxylic acid (TCA) cycle whose dysfunction underlies multiple pathological conditions. As a rate-limiting component of the multi-enzyme complex converting α-ketoglutarate to succinyl-CoA, OGDH plays a pivotal role in energy metabolism, with its impairment directly linked to neurodegenerative diseases like Alzheimer's and Parkinson's through mechanisms involving oxidative stress, bioenergetic failure, and aberrant glutamate signaling. The enzyme's susceptibility to reactive oxygen species makes it particularly vulnerable in ischemic conditions, contributing to reperfusion injury in stroke and myocardial infarction. In cancer biology, OGDH exhibits dual roles—while its downregulation promotes the Warburg effect in many tumors, certain malignancies demonstrate OGDH overexpression to sustain anabolic pathways. Metabolic disorders including type 2 diabetes show altered OGDH activity affecting glucose homeostasis, while inborn errors of OGDH function cause severe developmental delays and lactic acidosis. Recent evidence implicates OGDH in immune regulation through its influence on citrate metabolism and inflammatory signaling, suggesting therapeutic potential in autoimmune diseases. The enzyme's structural complexity, with its thiamine pyrophosphate cofactor requirement and multiple regulatory sites, presents unique opportunities for pharmacological modulation across these diverse disease contexts, though targeting specificity remains challenging due to its central metabolic position.

Background

This gene encodes one subunit of the 2-oxoglutarate dehydrogenase complex. This complex catalyzes the overall conversion of 2-oxoglutarate (alpha-ketoglutarate) to succinyl-CoA and CO(2) during the Krebs cycle. The protein is located in the mitochondrial matrix and uses thiamine pyrophosphate as a cofactor. A congenital deficiency in 2-oxoglutarate dehydrogenase activity is believed to lead to hypotonia, metabolic acidosis, and hyperlactatemia. Alternative splicing results in multiple transcript variants encoding distinct isoforms.

Interaction

The 2-oxoglutarate dehydrogenase complex is composed of OGDH (2-oxoglutarate dehydrogenase; E1), DLST (dihydrolipoamide succinyltransferase; E2) and DLD (dihydrolipoamide dehydrogenase; E3). It contains multiple copies of the three enzymatic components (E1, E2 and E3). In the nucleus, the 2-oxoglutarate dehydrogenase complex associates with KAT2A (PubMed:29211711).
