PIAS1

PIAS1 is a STAT1-specific repressor and a first member of the PIASs protein family when researchers screen for a protein that interacts with the C-terminal transcriptional activation domain of STAT1 by yeast two-hybrid. Its inhibition of STAT1, on the one hand, inhibits transcription by binding to dimeric STAT1 and masking the DNA binding domain of STAT1; on the other hand, it is achieved by binding to STAT1 monomer to hinder its dimerization. Recent studies have shown that PIAS proteins are not only inhibitors of STATS activation, they also interact with many other proteins, participate in the regulation of various transcription factors and a variety of protein modifications, thereby regulating cell proliferation, differentiation and apoptosis. It is closely related to neoplastic diseases. PIAS1 acts to inhibit inflammation through a variety of signal transduction pathways. In the mitogen-activated protein kinase signal transduction pathway, PIAS1 interacts with Net to cause SUMOylation to promote the transcriptional repression of Net and reduce its Ras activates the response of the signal and inhibits the inflammatory response. In addition, PIAS1 can also participate in the regulation of inflammatory responses by blocking the activation of STAT1, NF-kB and other signal transduction pathways.
