PRKACA: The Catalytic Engine of PKA Signaling
Protein Kinase CAMP-Activated Catalytic Subunit Alpha (PRKACA) is the most ubiquitously expressed catalytic subunit of Protein Kinase A. PKA is found as an inactive tetramer composed of two regulatory subunits and two catalytic subunits. In response to an increase in intracellular cAMP levels, cAMP binds to the regulatory subunits and this causes a conformational change that dissociates and activates the catalytic subunits such as PRKACA. The activated kinase phosphorylates hundreds of substrate proteins on serine and threonine residues to control diverse processes including metabolism, gene transcription, cell growth, and division. Activating mutations in PRKACA have been implicated as drivers of certain endocrine tumors.

Background
It phosphorylates various other proteins and substrates, thereby modifying their activity. Protein kinase A catalytic subunit (PKA Cα) belongs to the AGC kinase family (protein kinases A, G, and C) and plays a role in the regulation of cellular processes including glucose metabolism, cell cycle progression, and contextual memory.

A number of inactive tetrameric holoenzymes are produced by the combination of homo- or heterodimers of the different regulatory subunits associated with two catalytic subunits. cAMP causes the dissociation of the inactive holoenzyme into a dimer of regulatory subunits bound to four cAMP and two free monomeric catalytic subunits. The cAMP-dependent protein kinase catalytic subunit binds PJA2.
