PRKAR2A: An Anchoring Hub for PKA Signaling
Protein Kinase CAMP-Dependent Type II Regulatory Subunit Alpha (PRKAR2A) is one of four major regulatory subunits of the PKA holoenzyme. Like the other regulatory subunits, it functions to bind and inactivate the PKA catalytic subunit in a cAMP-dependent manner. The Type 2 subunits (R2A and R2B) differ from the Type 1 subunits in that they are themselves substrates for autophosphorylation by the catalytic subunit. More importantly for its role in determining PKA subcellular organization, the Type 2 regulatory subunits bind to A-Kinase Anchoring Proteins (AKAPs) and mediate PKA complex localization to defined subcellular compartments. By tethering PKA to its AKAPs, these anchoring proteins ensure that PKA is optimally positioned near its cognate substrates for maximal spatial and temporal control over cAMP signaling.

Background
cAMP is a second messenger important in many biological processes. cAMP mediates its actions through activation of cAMP-dependent protein kinase, which in turn transduces the signal by phosphorylating various target proteins. The inactive kinase holoenzyme is a tetramer of two regulatory and two catalytic subunits. cAMP causes dissociation of the inactive holoenzyme into a dimer of regulatory subunits each bound to 4 cAMP molecules and two free monomeric catalytic subunits. In humans, four distinct regulatory subunits and three catalytic subunits have been identified. The protein encoded by this gene is one of the regulatory subunits. This regulatory subunit can be phosphorylated by the activated catalytic subunit. It may bind to a variety of A-kinase anchoring proteins and may determine the subcellular localization of the cAMP-dependent protein kinase.

The inactive form of the enzyme is composed of two regulatory chains and two catalytic chains. Activation by cAMP produces two active catalytic monomers and a regulatory dimer that binds four cAMP molecules. Interacts with AKAP4 and CBFA2T3. Interacts with the phosphorylated form of PJA2. Interacts with MYRIP. This interaction may link PKA to components of the exocytosis machinery, thus facilitating exocytosis, including insulin release (By similarity). Forms a complex composed of PRKAR2A, GSK3B and GSKIP through GSKIP interaction; facilitates PKA-induced phosphorylation and regulates GSK3B activity (PubMed:27484798). Interacts with ADCY8; inhibits adenylate cyclase activity through PKA phosphorylation (PubMed:22976297).
