RCAN1: The Endogenous Brake on Calcineurin Signaling
Regulator of Calcineurin 1 (RCAN1), also known as DSC1, is a protein that in humans is encoded by the RCAN1 gene. The RCAN1 gene is on chromosome 21 in the Down syndrome critical region. RCAN1 is a potent endogenous feedback inhibitor of calcineurin. RCAN1 directly binds to the catalytic subunit of calcineurin and inhibits its phosphatase activity, thus preventing the dephosphorylation of NFAT and other substrates of calcineurin. RCAN1 expression is itself induced by calcineurin/NFAT signaling, and this constitutes a negative feedback loop. The chronic overexpression of RCAN1 by trisomy 21 is thought to underlie many features of Down syndrome, such as intellectual disability and early-onset Alzheimer's disease.

Background
The protein encoded by this gene interacts with calcineurin A and inhibits calcineurin-dependent signaling pathways, which may play a role in central nervous system development. This gene is located in the minimal candidate region for the Down syndrome phenotype, and it is overexpressed in the brain of Down syndrome fetuses. Chronic overexpression of this gene may lead to neurofibrillary tangles, such as those seen in Alzheimer disease. Alternative splicing results in multiple transcript variants.
