• The Collagen type II antibody has been reported in applications of Enzyme-linked Immunosorbent Assay, Surface Plasmon Resonance.
  Enzyme-linked Immunosorbent Assay: CII (1 μg/well) was coated to 96 Micro Well MaxiSorp™ Enzyme-linked Immunosorbent Assay plates (Nunc, Denmark) overnight at 4°C whereas the wells for blank controls were coated with 4‐μg BSA (bovine serum albumin). After coating, the wells were blocked with 2% BSA in phosphate buffered saline (PBS) for 1 h and subsequently incubated with the CIIC1scFv at different dilutions overnight. The His‐tag served for the detection of bound CIIC1scFv by an unlabeled Penta‐His Antibody (Qiagen), diluted 1:500 in PBS pH 7.4 and incubated for 2 h at room temperature.
  SPR: Surface Plasmon Resonance measurements were performed on a BIAcore 2000 instrument. CII (100 μg/mL) diluted in 10 mM acetate buffer, pH 4.5 dissolved in running buffer (HBS‐EP [0.01 M HEPES, pH 7.4, 0.15 M NaCl, 3 mM EDTA, 0.005% Surfactant P20]) was covalently immobilized on the CM5 sensor chip at a density of 600 response units by using standard amine‐coupling chemistry.
