Human Anti-HNE Recombinant Antibody (clone R310) (CAT#: PABL-197)
Recombinant Human Antibody (R310) is capable of binding to HNE, expressed in HEK 293 cells. Expressed as the combination of a heavy chain (HC) containing VH from anti-HNE mAb and CH1-3 region of human IgG1 and a light chain (LC) encoding VL from anti-HNE mAb and CL of human kappa light chain. Exists as a disulfide linked dimer of the HC and LC hetero-dimer under non-reducing condition. This antibody showed only a slight crossreactivity with the native double-stranded DNA, whereas the Ab immunoreactivity was dramatically enhanced by the treatment of the DNA with 4-oxo-2-nonenal (ONE), an analog of HNE.

A coating antigen was prepared by incubating calf-thymus DNA (1 mgml) with 5 mM aldehydes in 1 ml of 0.1 M sodium phosphate buffer (pH 7.4) for 24 h at 37°C. One microgram of antigen (calf-thymus DNA or oligonucleoside 50-mers) was coated per well in polystyrene plates, and Ab binding was detected.
