The indicated HTNV-specific MAbs, HTNV-specific mouse hyperimmune ascitic fluid (HTN-poly), or HPS convalescent-phase serum from Argentina (AND-poly) was used to immunoprecipitate proteins from COS cells transfected with pWRG/HTN-M(x) (H) or pWRG/AND-M (A). +, G1 or G2 was immunoprecipitated; −, no protein was immunoprecipitated; *, both G1 and G2 were immunoprecipitated.

COS cells were transfected with pWRG/HTN-M or a negative control plasmid (pWRG7077) and, after 24 h, radiolabeled cell lysates were prepared for analysis by RIPA. Expression products were immunoprecipitated with a polyclonal mouse hyperimmune ascitic fluid against HTNV (HTN HMAF), a G1-specific MAb (MAb 6D4), or a G2-specific MAb (MAb 23G10).

Radiolabeled HTNV proteins were immunoprecipitated with HFRS patient serum (lane HP) or with recombinant human antibodies as indicated above each lane. The positions of the viral envelope glycoproteins, G1 and G2, and the nucleocapsid protein, N, are indicated to the left of the autoradiogram.

FITC conjugated anti-human IgG Fc antibodies were used for the detection. Cell culture supernatants of SF9 cells infected with recombinant baculoviruses expressing human IgG antibodies to G1 and N were incubated with the slides at 378C for 30 min. Bound antibodies were detected with FITC conjugated anti-human human Fc antibodies.
