  • Peptide linkers, belonging to Enzymatically cleavable linkers, combine greater systemic stability with rapid enzymatic release of the drug in the target cell. The scission of peptidic bonds relies on lysosomal proteolytic enzymes, which have very low activities in blood due to endogenous inhibitors and the unfavorably high pH value of blood.

  • Auristatins are water-soluble dolastatin analogs of dolastatin 10. Dolastatin 10 belongs to dolastatin family and it can powerfully bind to tubulin, thus inhibiting polymerization mediated through the binding to the vinca alkaloid binding domain, and causes cell to accumulate in metaphase arrest.

Since Protein A binds to many IgG isotypes, this product was incredibly versatile for our lab. We could mix it with various antibodies to create instant ADCs for preliminary testing. The vc linker and MMAE payload worked consistently across different setups.

Efficient Tubulin Inhibition Assay
We used this conjugate to test the internalization of several unknown antibodies. The MMAE toxin effectively depolymerized microtubules in cells where the primary antibody internalized, giving us a clear and quick functional readout for our candidate screening.

  1. What is the advantage of using Protein A in this conjugate?

    A: Protein A binds to the Fc region of immunoglobulins from many species. By conjugating Protein A to MMAE, this product can be mixed with various primary antibodies to form non-covalent ADCs, facilitating the rapid screening of antibody internalization and cytotoxicity.

  2. How is the MMAE toxin released from the Protein A conjugate?

    A: The MMAE is attached via a vc (valine-citrulline) linker. This is an enzymatically cleavable peptide linker. Upon internalization of the Protein A-antibody complex into the lysosome, proteases cleave the linker, releasing the active toxin.
