This research investigates the therapeutic potential of microcystin-LR, an environmental cyanobacterial toxin, in treating idiopathic pulmonary fibrosis (IPF). The study demonstrates that chronic oral administration of microcystin-LR attenuates pulmonary fibrosis in both bleomycin-induced rat and fluorescein isothiocyanate-induced mouse models. Mechanistically, microcystin-LR reduces TGF-β1/Smad signaling in pulmonary tissues and blocks epithelial-mesenchymal transition (EMT) and fibroblast-myofibroblast transition (FMT) by suppressing the differentiation of CD206+ M2-like macrophages. Further investigation revealed that microcystin-LR binds to glucose-regulated protein 78 kDa (GRP78) and suppresses endoplasmic reticulum unfolded protein response (UPRER) signaling pathways, which modulates M2 polarization of macrophages and contributes to the alleviation of pulmonary fibrosis.
Creative Biolabs provided two independent microcystin-LR antibodies (CAT#: MOB-647) that were crucial for the immunoprecipitation and protein mass spectrometry experiments identifying GRP78 as the binding partner of microcystin-LR. This finding was central to revealing the novel mechanism through which microcystin-LR exerts its anti-fibrotic effects by modulating ER stress and M2 macrophage polarization, opening potential new therapeutic approaches for treating IPF, a condition with currently limited treatment options.
