Anti-BCL6 Antibody Prodrug, Protease Activated (GN15)

The Anti-BCL6 Antibody Prodrug, Protease Activated (GN15) is an antibody prodrug comprised of a recombinant human antibody targeting human BCL6, linked to a masking peptide that conceals the active antigen-binding site via a protease-cleavable linker. The mask peptide is recombinantly attached to the amino terminus of either the light or heavy chain of the antibody, designed to block access to the antigen-binding site and physically prevent the antibody from binding to BCL6 protein. Once the masking peptide is cleaved by matrix metalloproteinases (MMPs), the prodrug is activated, allowing the unmasked monoclonal antibody component to bind to BCL6 effectively. BCL6 (B-cell Lymphoma 6) is a transcription factor that acts as a key oncogene in many types of B-cell lymphomas, particularly diffuse large B-cell lymphoma (DLBCL) and follicular lymphoma. BCL6 prevents B-cell differentiation and promotes an aggressive, proliferative phenotype.

- The protein encoded by this gene is a zinc finger transcription factor and contains an N-terminal POZ domain. This protein acts as a sequence-specific repressor of transcription, and has been shown to modulate the transcription of STAT-dependent IL-4 responses of B cells. This protein can interact with a variety of POZ-containing proteins that function as transcription corepressors. This gene is found to be frequently translocated and hypermutated in diffuse large-cell lymphoma (DLCL), and may be involved in the pathogenesis of DLCL. Alternatively spliced transcript variants encoding different protein isoforms have been found for this gene.
