Competition of purified Fabs with ch14.18 on GD2. GK2 and GK8 Fabs were purified by affinity chromatography using immobilized anti-human Fab Abs. Competition of purified Fabs with ch14.18 on GD2 clearly showed that GK2 Fab and GK8 Fab were able to significantly inhibit (GK2 Fab) or completely suppress binding (GK8 Fab) of ch14.18 to GD2 even at low concentrations, in contrast with an anti-BSA-Fab (negative control). Depicted is the percentage of binding inhibition as determined by B/Bo, where B is absorbance with inhibitor and Bo is absorbance without inhibitor. Bo was set at measurement end point OD of 2.5.

Competition of purified Fabs with the natural anti-idiotypic Abs of patient 1 on 14G2a. Purified GK2 and GK8 Fabs competed with the natural anti-idiotypic Abs of patient 1, i.e., diluted patient's serum, for binding to 14G2a. GK8 Fab demonstrated nearly complete binding inhibition, indicating that it represents the majority of the patient's anti-idiotypic Abs. The patient's serum had to be diluted to adjust for comparable IgG concentrations of both competitors.
