The anti-Integrin α2β1 pH-dependent antibody (VS-0825-YC363) targets human Integrin α2β1 in a pH-dependent manner, with its affinity remaining high at neutral pH but weakening at a lower pH, such as within the acidic environment of endosomes. The anti-Integrin α2β1 acid-switched antibody was engineered by incorporating histidine residues into its variable regions. The introduction of histidine substitutions within its Complementarity-Determining Regions (CDRs) enables pH-responsive antigen binding, which leads to enhanced Integrin α2β1 clearance. The antibody operates in a cyclical manner, binding Integrin α2β1 in the extracellular fluid and releasing it in acidified endosomes. Integrin Integrin α2β1 is a cell surface receptor for collagen and laminin. It is involved in cell adhesion, migration, and differentiation, playing a key role in wound healing, fibrosis, and cancer progression.
