The anti-Integrin αvβ8 pH-dependent antibody (VS-0825-YC367) targets both human and mouse Integrin αvβ8 in a pH-dependent manner, with its affinity remaining high at neutral pH but weakening at a lower pH, such as within the acidic environment of endosomes. The anti-Integrin αvβ8 acid-switched antibody was engineered by incorporating histidine residues into its variable regions. The introduction of histidine substitutions within its Complementarity-Determining Regions (CDRs) enables pH-responsive antigen binding, which leads to enhanced Integrin αvβ8 clearance. The antibody operates in a cyclical manner, binding Integrin αvβ8 in the extracellular fluid and releasing it in acidified endosomes. Integrin αvβ8 is a receptor for extracellular matrix proteins and a key activator of latent TGF-β. It is involved in nervous system development and its dysregulation is linked to certain cancers.
