The anti-MMP7 pH-dependent antibody (VS-0825-YC368) is a recombinant monoclonal antibody that exhibits pH-dependent antigen binding to human MMP7. By introducing histidine mutations that act as a pH-switch, the antibody demonstrates a preferential affinity for MMP7 under acidic conditions compared to physiological pH. Using a yeast surface display system with a pH-dependent screening pressure, this mAb is identified which shows a significant increase in dissociation from MMP7 at pH 6.0. This pH-sensitive antibody retains its binding affinity on tumor cells while exhibiting negligible binding to the receptor on normal cells at physiological pH. Human MMP7, also known as Matrilysin, is a protease that degrades components of the extracellular matrix. It is involved in tissue remodeling and is frequently overexpressed in various cancers, promoting tumor invasion and metastasis.
