The anti-PROK2 eliminating antibody is an engineered pH-sensitive antibody against human PROK2. PROK2, expressed in the circadian clock, acts as a chemoattractant and is linked to Kallmann syndrome. The antibody has a novel PROK2-sweeping activity in vivo, which selectively eliminatesthe soluble PROK2 from plasma. Compared to conventional antibody, the mAb exerted in vivo efficacy by simultaneously engineering both pH-dependent antigen binding, to mimic the pH-dependent receptor-ligand binding of such endocytic receptors, and increased FcRn binding affinity at neutral pH, to mimic the cell-bound form of the receptors.
