The gold antibody labeling kit (Maleimide) allows for easy covalent labeling of biomolecules with gold. The procedure is quick, with a 5-minute hands-on time and the conjugate is ready within 1 hour. The proprietary surface coat with maleimide groups ensures stability even under extreme conditions. Purified antibodies in suitable buffer at the right concentration are recommended for labeling. Consider using antibody purification and concentration kits if needed.

- Procedure Steps
- Equilibrate all materials and prepared reagents to room temperature and gently stir before use.
4.1 Dilute the biomolecule in diluent to a final volume of 40 µL (3x 2.5 µg kit) or 400 µL (1x 25 µg kit).
4.2 Add 5 µL (3x 2.5 µg kit) or 50 µL (1x 25 µg kit) reaction buffer. Mix gently.
4.3 Remove the cap from the Gold Mix vial, mix and pipette the sample (biomolecule in reaction buffer) directly onto the lyophilized material. Resuspend by gently aspirating and redispensing the liquid once or twice with a pipette.
4.4 Place the cap back on the vial and place it at room temperature (20-25°C); a 30 minute incubation is recommended for proteins or antibodies and a 60 minute incubation is recommended for oligonucleotides; longer incubation times have no negative effects on the conjugate.
4.5 After 30 minutes (or longer) of incubation, add 5 µL (3x 2.5 µg kit) or 50 µL (1x 25 µg kit) of quencher (supplied as a lyophilized product and should be reconstituted in 100 µL of water; store remaining quencher at -20°C after use).
The conjugate is ready for use after 20 minutes. You now have 50 µL (3x 2.5 µg kit) or 500 µL (1x 25 µg kit) of 20 OD of conjugate.
4.6 Determine the concentration of the conjugate: To determine the effective concentration of the conjugate obtained, we recommend measuring the maximum absorbance of light using a UV-Vis spectrophotometer after diluting the sample to the range appropriate for your equipment.

- Upon receipt, store the kit in a dark place at a temperature of -20°C immediately. Check below for storage instructions for individual components. The kit can be stored for up to 1 year from receipt if the components have not been reconstituted. Ensure to avoid repeated freeze-thaw cycles of reagents.
- Conjugate Storage
- Storage at 4°C is advised for all conjugates. Long-term storage may benefit from the addition of a preservative. The optimal storage conditions for specific conjugates should be determined through experimentation.
