- Procedure Steps
- 1. Allow all reagents to warm to room temperature.
2. Dilute the stock antibody to a appropriate concentration using the antibody diluent provided in the kit.
3. For each reaction, add reaction buffer and the now diluted antibody according to the table below and mix gently.
4. Transfer 45 μL of the mixture to the gold reagent vial if using the 3 or 10x 1 µg kits or 450 μL of the mixture to the gold reagent vial if using the larger volume kit (1x 10 µg). Dissolve the gold reagent by gently pipetting up and down. Let the reaction sit at room temperature for 15 minutes.
5. After 15 minutes, stop the reaction by adding the appropriate volume of Gold quencher according to the table (Contact Us). Mix well but gently.
6. Let the reaction sit for 5 minutes. You now have 20 OD of conjugate (50 μL for the 3 and 10x 1 µg kits, 500 μL for the 1x 10 µg kit). Dilute further as needed for your application.
7. Storage of conjugate antibody:
After antibody labeling, we recommend storing the conjugate at +4°C. The gold quencher added in the last step of conjugation is a good storage buffer for the conjugate. Do not store the conjugate at -20°C.
The overall stability of the conjugate will be determined by the stability of the antibody itself, as it will degrade first.
As long as your antibody is stable, the conjugate will also be stable.
8. Measuring the concentration of the conjugate:
To determine the effective concentration of the conjugate obtained, we recommend measuring the maximum absorbance using a UV-Vis spectrophotometer after diluting the sample to a range that is appropriate for your equipment (e.g., if the conjugate is 20 OD and the dilution is 1: 20, the maximum absorbance for a 1 cm pathlength would be expected to be approximately 1 AU).

2. Buffer components & conditions should not include the following:
PBS / Thimerosal / Thimerosal / Merthiolate / BSA / Gelatin / Tris / Glycine / Carboxylic acids (e.g. EDTA, Citrate) / Nucleophilic components (Primary amines e.g. amino acids or ethanolamine and thiols e.g. mercaptoethanol or DTT)
Note: Individually, these components should not affect the reaction. In combination, however, the reaction may be affected.

- Kit Storage
- Upon receipt of the kit, please store it at -20°C. The quencher, reaction buffer, and antibody diluent may be stored at either 4°C or -20°C.
- Conjugate Storage
- After antibody labeling, we recommend storing the conjugate at +4°C. Gold quencher added in the final step of conjugation is a good storage buffer for the conjugate. Do not store the conjugate at -20°C.
