- Procedure Steps
- After the HRP-antibody conjugation reaction, the success of the conjugation can be visually checked by running the sample on strips with the following steps:
1. Dilute the 10x running buffer with distilled water to make it 1x concentration.
2. Add 0.01% BSA to the 1x running buffer to create a final concentration.
3. Dilute the HRP-antibody conjugate in 1x running buffer with 0.01% BSA.
4. Load 40 µL of the diluted conjugate into a 96-well non-sticky plate or a suitable container.
5. Insert the strip into the well with the nitrocellulose membrane dipped into the diluted conjugate.
6. Run for 10 minutes.
7. Add 40 µL of HRP detection solution to each well.
8. Run for another 15 minutes.
9. Check the result visually.
The kit also includes a positive control consisting of lyophilized GAR-HRP, which should be run separately on a strip:
10. Reconstitute the lyophilized positive control in 50 μL dH2O to obtain a concentration of 10 μg/mL.
11. Dilute the reconstituted control to a concentration of 100 ng/mL by adding 4 µL to 396 µL of 1x running buffer + 0.01% BSA.
12. Dilute the 100 ng/mL control to 1ng/mL by adding 4 µL to 396 µL of 1x running buffer + 0.01% BSA.
13. Load 40 µL of the diluted positive control into each well.
14. Insert the strip into the well.
15. Run for 10 minutes.
16. Add 40 µL of HRP detection solution to each well.
17. Run for another 15 minutes.
18. Check the result visually.

- Upon arrival, please store Protein A/G Strips and the HRP detection solution at +4°C, and all other components at -20°C. The kit can be stored for up to 1 year from the date of receipt, as long as the components have not been reconstituted.

- Fluorescent Dye Introduction
