The streptavidin antibody conjugation check kit is a convenient and cost-effective tool for confirming the successful conjugation of streptavidin to antibodies. The kit utilizes a nitrocellulose membrane with immobilized Protein A and Protein G to capture the streptavidin-antibody conjugate, along with 40 nm gold-conjugated Biotin for detection. This allows for a visible red line to appear on the strip, indicating successful conjugation. No specialized equipment is required, making it a quick and simple test to perform.

- Procedure Steps
- Before starting, make sure all materials and reagents are at room temperature and gently mix them.
1. Dilute the 10x running buffer with distilled water to make a 1x solution.
2. Mix 0.1% BSA into the 1x running buffer.
3. Reconstitute the 40 nm gold biotin by adding 150 μL of 1x running buffer + 0.1% BSA to achieve a concentration of 2 OD.
4. Dilute your samples in 1x running buffer + 0.1% BSA; use 10 µL for each test with a range of 250-15.6 ng/mL.
It is recommended to run samples in duplicate and include a positive control.
5. For a single test, combine 10 µL of diluted streptavidin-antibody, 4 µL of reconstituted Gold Biotin 2 OD, and 26 µL of 1x running buffer + 0.1% BSA, mix well.
6. Dispense 40 µL of the mix into each well of a non-sticky 96-well plate.
7. Insert the strip into the well.
8. Run the test for 10 minutes and visually inspect for the appearance of a red line indicating successful conjugation.
9. Reconstitute the vial with 300 μL of 1x running buffer + 0.1% BSA.
10. For the positive control, mix 10 µL of control, 4 µL of reconstituted Gold Biotin 2 OD, and 26 µL of 1x running buffer + 0.1% BSA.
11. Load 40 µL of the positive control mix into each well.
12. Insert the strip into the well.
13. Run the test for 10 minutes.

- Upon arrival, please store Protein A/G Strips and the HRP detection solution at +4°C, and all other components at -20°C. The kit can be stored for up to 1 year from the date of receipt, as long as the components have not been reconstituted.
