Our antibody labeling kit is specifically designed for the conjugation of MMAF with a releasable VC-PAB linkage to antibodies (IgG). This kit contains maleimide-activated MMAF (Mc-VC-PAB-MMAF) which can be directly coupled to the antibodies after reduction through alkylation in a single step.
The reduced thiols at the heavy chains of the antibody serve as the labeling sites for conjugation with MMAF. The VC-PAB linkage is designed to be cleavable by Cathepsin B, allowing for the release of MMAF from the antibody once it reaches its target.
This antibody labeling kit provides a convenient and efficient way to attach MMAF to antibodies for targeted drug delivery applications.

- 1) A 1.5 mL microcentrifuge tube with less than 1 mg of hazardous MC-VC-PAB-MMAF solution.
2) A 0.5 mL microcentrifuge tube containing less than 10 mg of non-hazardous Reagent A.
3) A tube holding either 1.5 or 6 mL of non-hazardous solution A.
4) A bottle with either 4 or 12 mL of non-hazardous reducing buffer.
5) A bottle with either 4 or 12 mL of non-hazardous labeling buffer.
6) A plastic bottle containing 20 or 60 mL of non-hazardous Storage buffer (phosphate buffer saline).
7) A plastic bag containing centrifugal filter devices, desalting columns, and microcentrifuge tubes that are non-hazardous.

- Kit Storage
- Upon receiving the items, please store Box 1 in a freezer at -20°C or below and remove from the package. Store Box 2 in a refrigerator at 2-8°C.

- The total amount of protein should ideally be between 1-3 mg, as measured by UV, with a purity of greater than 90% determined by gel electrophoresis.

- Fluorescent Dye Introduction
- Monomethyl auristatin F (MMAF) with the Mc-VC-PAB linker is a potent cytotoxic compound that inhibits cell division by blocking tubulin polymerization, leading to the disruption of microtubule dynamics. This mechanism of action allows MMAF to effectively target and kill rapidly dividing cancer cells.
The addition of the Mc-VC-PAB linker further enhances the specificity and efficacy of MMAF by allowing for targeted delivery to tumor cells. The linker is stable in extracellular fluid but is cleaved by cathepsin B once inside the tumor cell, activating the antimitotic mechanism and increasing the potency of MMAF against cancer cells.
In addition to its anticancer activity, MMAF also exhibits antioxidant, anti-inflammatory, and insecticidal activities, making it a versatile compound with a range of potential therapeutic applications.
