Background
Monomethyl auristatin F (MMAF) with the Mc-VC-PAB linker is a potent cytotoxic compound that inhibits cell division by blocking tubulin polymerization, leading to the disruption of microtubule dynamics. This mechanism of action allows MMAF to effectively target and kill rapidly dividing cancer cells.

The linker is stable in extracellular fluid but is cleaved by cathepsin B once inside the tumor cell, activating the antimitotic mechanism and increasing the potency of MMAF against cancer cells.
