  • Procedure Steps
  • 1. Oligonucleotide activation
    Add 100 μL of oligonucleotide to the oligonucleotide activation reagent bottle.
    Mix gently and incubate at room temperature for 30 minutes.
    During the incubation, proceed to 2.
    2. Antibody activation
    Add 100 μL of antibody (at a concentration of 1 mg/mL) to the antibody activation Reagent bottle. Mix gently and incubate at room temperature for 30 minutes.
    During the incubation, proceed to 3.
    3. Active desalting
    Note: Use one column per sample to be desalted.
    Columns are designed for single use. Discard after use.
    3.1 Secure each separation column in a vertical position.
    Remove the top cap first. Then remove the bottom cap and allow the storage solution to flow through the column. Discard the flow-through from the column.
    3.2 Equilibrate each column by adding 3 mL of wash buffer to the top of the column and allowing the liquid to flow through by gravity. Discard the flow-through. Repeat 4 more times.
    3.3 After 30 minutes of incubation in 4.1 and 4.2 for activation, add 100 μL of activated oligonucleotide or antibody to the top of the column and allow the liquid to completely absorb into the column. Collect the flow-through and save until successful binding is confirmed.
    3.4 Add 550 μL of wash buffer to the top of the column. This will push the activated material to the bottom of the column. Allow this liquid to completely absorb before proceeding to the next step. Collect the flow-through and save until successful binding is confirmed.
    3.5 Place a clean microcentrifuge tube under the column. Add 300 µL of wash buffer to the top of the column.
    3.6 Collect the eluate from the column. This column eluate (300 µL) contains the activated oligonucleotide or antibody that can be used immediately for binding.
    4. Storage of activated materials
    Oligonucleotides and control oligonucleotides
    Activated oligonucleotides can be stored at room temperature for up to 8 hours. For long-term storage of up to 12 months, storage at -20°C is recommended.
    Antibodies and control antibodies:
    Activated antibodies should be stored on ice and used within 2 hours. Activated antibodies are not stable enough for long-term storage.
    5. Generation of purified antibody-oligo conjugates
    This kit can be used to generate antibody-oligo conjugates with a range of antibody: oligo ratios. Simply add varying amounts of oligo to the antibody as described in the table below. The preferred ratio will depend on the experiment in which the conjugate will be used and may need to be determined experimentally.
    5.1 Add 300 μL of activated antibody to an appropriate volume of activated oligo and wash buffer (contact us).
    5.2 Mix and incubate at room temperature for 1 hour.
    The conjugate may also be incubated overnight at room temperature with no adverse effects.
    5.3 Your conjugate is now ready. If your application requires, you may also purify the conjugate to remove any unbound oligo.
    5.4 Any unused activated oligo may be stored at -20°C.

  • Upon receipt, store the activation reagent and buffer packs at 4°C and the conjugate cleanup reagent and separation columns at room temperature.
  • Conjugate Storage
  • To maximize stability, we recommend storing conjugates at low temperatures in the most concentrated form possible. We recommend checking with antibody and oligonucleotide manufacturers if their products can be stored in 50% glycerol at -20°C-you should be able to store most conjugates under these conditions as long as they are compatible with unconjugated antibodies and oligonucleotides. Adding a preservative may also be helpful if appropriate for your reagents and subsequent experiments.
