AHR: A Ligand-Activated Sensor Bridging Environment and Immunity

The Aryl Hydrocarbon Receptor (AHR) is a ligand-activated transcription factor that is localized to the cytosol. Long known for its role in mediating the toxic effects of environmental pollutants like dioxins, it has become apparent that AHR is also capable of binding a large number of endogenous ligands, including metabolites of tryptophan. In this way, AHR is a critical sensor of the tissue microenvironment. AHR signaling has profound effects on the differentiation of immune cell subsets, especially T cells (Th17/Treg balance) and innate lymphoid cells. It is frequently dysregulated in various cancers and autoimmune diseases and is an exciting therapeutic target.

Background

The protein encoded by this gene is a ligand-activated helix-loop-helix transcription factor involved in the regulation of biological responses to planar aromatic hydrocarbons. This receptor has been shown to regulate xenobiotic-metabolizing enzymes such as cytochrome P450. Before ligand binding, the encoded protein is sequestered in the cytoplasm; upon ligand binding, this protein moves to the nucleus and stimulates transcription of target genes. Binds MYBBP1A (By similarity). Interacts with coactivators including SRC-1, RIP140 and NOCA7, and with the corepressor SMRT (PubMed:10395741).
