ALAS1: A Key Regulator in Hepatic Metabolic Disorders and Porphyria Pathogenesis

ALAS1, the rate-limiting enzyme in hepatic heme biosynthesis, plays a critical role in systemic metabolic disorders. Dysregulation leads to acute hepatic porphyrias, characterized by neurovisceral attacks triggered by accumulated neurotoxic porphyrin precursors (ALA/PBG). Impaired activity disrupts cytochrome P450 function, exacerbating drug metabolism defects and hormonal imbalances. Recent studies link its aberrant expression to non-alcoholic fatty liver disease progression through mitochondrial dysfunction and oxidative stress. In certain cancers, elevated activity supports tumor proliferation by fueling heme-dependent oxygen sensing pathways. The enzyme’s nutritional regulation (B6-dependency) creates therapeutic vulnerabilities for metabolic modulation. Its tissue-specific isoforms offer diagnostic value for distinguishing hepatic vs. erythroid pathologies. Current research explores small-molecule inhibitors to attenuate overproduction of toxic intermediates while maintaining basal heme levels, presenting a promising approach for managing acute porphyria crises and metabolic comorbidities.

Background

This gene encodes the mitochondrial enzyme which is catalyzes the rate-limiting step in heme (iron-protoporphyrin) biosynthesis. The enzyme encoded by this gene is the housekeeping enzyme; a separate gene encodes a form of the enzyme that is specific for erythroid tissue. The level of the mature encoded protein is regulated by heme: high levels of heme down-regulate the mature enzyme in mitochondria while low heme levels up-regulate. A pseudogene of this gene is located on chromosome 12. Alternative splicing results in multiple transcript variants encoding different isoforms.

Rabbit Anti-ALAS1 Recombinant Antibody (clone 2A9) (CAT#: ZG-0541J)
This Rabbit Anti-ALAS1 Recombinant Antibody worked well in ELISA using HEK293 cell lysates. Showed specific binding with low background. Also tested on mouse liver tissue for Western Blot, giving clear bands. Easy to handle, consistent results across assays. Good for detecting ALAS1 in different sample types without issues.

Mouse Anti-ALAS1 works well for WB in human liver and HepG2 lysates, showing a clean ~70kDa band at 1:800 dilution. The signal is specific but requires 5% BSA blocking to reduce background in tissue samples. While it reliably detects overexpression, endogenous levels in primary hepatocytes need longer exposure. No cross-reactivity with ALAS2 observed. Consistent performance across multiple batches makes it suitable for hepatic metabolism studies, though low-abundance samples may need optimization.

In our experience with the VS3-CJ627 anti-ALAS1 antibody, it's proven effective for immunofluorescence studies in both cultured hepatocytes and frozen liver tissue sections. The antibody reliably detects endogenous protein at 1:300 dilution, showing expected mitochondrial staining pattern when combined with organelle markers. While the signal remains robust in acute porphyria patient samples, we noticed slight non-specific nuclear staining in cirrhotic liver tissues that required additional blocking. The antibody works particularly well for comparative studies of ALAS1 expression changes under metabolic stress conditions, though users should be aware that optimal fixation time varies between cell types.
